How do you tweak the settings so that its possible to define a glass opbject in the cam locate. And what colour background is best for it?

The wrist camera is not designed to detect transparent object. You may not managed to detect glass objects.

Maybe you can adjust lighting to increase light reflection on the edges of the piece of glass to ease detection ?

Indeed it can be hard to detect glass part. In the same idea as @bcastets increasing the reflection on your part might be helpful. You could also have a dark non reflective background and increase the lighting AND the exposure time. It will make the picture brighter and hopefully give you a good edge difference with the background.
